Functional enhancement of exosomes derived from NK cells by IL-15 and IL-21 synergy against hepatocellular carcinoma cells: The cytotoxicity and apoptosis in vitro study

چکیده
Exosomes are released by various cells, including natural killer (NK) cells and transport signaling molecules for the intercellular communication. Hepatocellular carcinoma (HCC), also known as primary liver cancer, is often inoperable difficult to accurate diagnosis. Notably, prognosis underlying mechanisms of HCC not fully understood. Exosomes-derived NK (NK-exos) express unique cytotoxic proteins with a killing ability in tumors can easily penetrate tumor tissues improve their targeting ability. cell functions, inducing cellular cytotoxicity modulated cytokines such interleukin (IL)-15 IL-21. However, effects cytokines-stimulated NK-exos treatment HCC, well known. In this study, we aimed investigate synergistic anti-tumor stimulated IL-15 IL-21 (NK-exosIL−15/21) Hep3B cells. Our findings revealed that NK-exosIL−15/21 expressed (perforin granzyme B) contained typical exosome markers (CD9 CD63) within size range 100–150 nm. Moreover, demonstrated induced enhancement apoptotic activity activating specific pro-apoptotic (Bax, cleaved caspase 3, PARP, perforin, inhibiting anti-apoptotic protein (Bcl-2). summary, our results suggest regulate strong increasing apoptosis through activation molecules.

Lactobacillus Acidophilus Cytotoxicity Effect and Apoptosis in Human Bladder Carcinoma Cells: An In Vitro Study
Introduction: Anticancer effects of Lactobacillus acidophilus, as a probiotic bacterium, have been indicated in several studies. There are common therapeutic options for bladder cancer treatment; however, their side effects and recurrence of disease are considerable. Therefore, complementary medication is essential and it must be safe and effective. In the present research, we assessed the anti...
متن کاملCytokine production and killer activity of NK/T-NK cells derived with IL-2, IL-15, or the combination of IL-12 and IL-18.
NK cell populations were derived from murine splenocytes stimulated by IL-2, IL-15, or the combination of IL-12 and IL-18. Whereas NK cells derived with the latter cytokines consisted of an homogeneous population of NK cells (DX5+CD3-), those derived with IL-2 or IL-15 belonged to two different populations, namely NK cells (DX5+CD3-) and T-NK cells (DX5+CD3+). Among NK cells, only those derived...
متن کاملDownregulation of Kinesin Spindle Protein Inhibits Proliferation, Induces Apoptosis and Increases Chemosensitivity in Hepatocellular Carcinoma Cells
Background: Kinesin spindle protein (KSP) plays a critical role in mitosis. Inhibition of KSP function leads to cell cycle arrest at mitosis and ultimately to cell death. The aim of this study was to suppress KSP expression by specific small-interfering RNA (siRNA) in Hep3B cells and evaluate its anti-tumor activity. Methods: Three siRNA targeting KSP (KSP-siRNA #1-3) and one mismatched-siRNA (...
